Detailed Introduction
Hygromycin B is an aminoglycoside antibiotic isolated from Streptomyces hygroscopicus . It inhibits protein synthesis by mistranslating mRNA templates, thereby killing bacteria, fungi, and eukaryotic cells. The gene hyg or hph can provide hygromycin B resistance to cells expressing the gene.
In Escherichia coli, the recommended selection concentration range of hygromycin B is 50-100 μg/ml. LB plates containing hygromycin B (yeast extract 5 g/l, Tryptone 10 g/l, NaCl 5 g/l, Agar 15 g/l, pH 7.5) can be stored at 4°C for 1 month. In mammalian cells, the recommended selection concentration range of hygromycin B is 100-800 μg/ml, which should be optimized for each cell line. The onset time of hygromycin B is longer than that of G418. Depending on the cell type, it usually takes 10 days to 3 weeks of culture to obtain a stable cell line.
