G418 is a kind of An aminoglycoside antibiotic produced by rhodorangea , its structure is similar to gentamicin . It inhibits protein synthesis in prokaryotic and eukaryotic cells by binding to ribosomes, leading to cell death. The neomycin resistance gene ( neo ) encodes APH3'II , which can make cells expressing this gene resistant to G418 .
In Escherichia coli , the recommended selection concentration range of G418 is 100-200 μg /ml . In mammalian cells, the selection concentration range of G418 is 400-800 μg/ml and should be optimized for each cell line.
