background:
Human epidermal growth factor receptor 2 (Her-2) is a transmembrane glycoprotein with receptor tyrosine kinase (RTK) activity encoded by the proto-oncogene CerbB2 (Her2/neu) . It belongs to the epidermal growth factor receptor family, which includes HER1 (erbB1, EGFR), HER2 (erbB2, NEU), HER3 (erbB3) and HER4 (erbB4). Studies have shown that ErbB2/HER2 has gene amplification and protein overexpression in 20% to 30% of primary invasive ductal breast carcinoma. Trastuzumab and Pertuzumab are both monoclonal antibodies that bind to the extracellular domain of ErbB2/HER2, but the two have different binding sites. Pertuzumab binds to the extracellular domain II of the ErbB2/HER2 receptor, while trastuzumab binds to the extracellular domain IV of the HER2 receptor.
