background:
ILDR2 belongs to the immunoglobulin superfamily. It may be involved in lipid homeostasis and endoplasmic reticulum stress pathways. ILDR2 is a novel B7-like protein with potent T-cell suppressive activity expressed on immune cells and in immune-privileged and inflamed tissues. A fusion protein consisting of the ILDR2 extracellular domain and the Fc fragment, bound to its putative counterpart on activated T cells, showed beneficial effects in a collagen-induced arthritis model and abolished the production of proinflammatory cytokines and chemokines in autologous synovial-like co-cultures of macrophages and cytokine-stimulated T cells. Collectively, these findings suggest that ILDR2 is a novel negative regulator of T cells with a potential role in the development of immune-related diseases, including autoimmunity and cancer.
