Prostate-specific membrane antigen (PSMA), also known as folate hydrolase 1 (FOLH1), glutamate carboxypeptidase 2 (GCP2), N-acetylated-α-linked acid dipeptidase I (NAALAD1), belongs to the M28 family of peptidases and the M28B subfamily. FOLH1/PSMA is stable at pH greater than 6.5. FOLH1/PSMA is a type II transmembrane zinc metallopeptidase with highest expression in the nervous system, prostate, kidney, and small intestine. FOLH1/GCP-2 is a homodimer, binding 2 zinc ions per subunit , and is required for NAALADase activity. The catalytic activity of PSMA involves the release of an unsubstituted C- terminal glutamyl residue, typically from Ac-Asp-Glu or poly -γ-glutamate. FOLH1/GCP-2/PSMA has folate hydrolase and N-acetylated-α-linked-acid dipeptidase ( NAALADase ) activities, with a preference for tri-α-glutamic acid peptides. GCP-2/PSMA is involved in prostate tumor progression and also exhibits dipeptidyl peptidase type IV activity in vitro, cleaving Gly-Pro-AMC.
