background:
dickkopf -related protein family (DKK-1, -2, -3, and -4) are secreted proteins with two cysteine-rich domains separated by a linker region. DKK1 participates in embryonic development by inhibiting the WNT signaling pathway, binds to LRP6 with high affinity, and prevents the formation of the Frizzled-Wnt-LRP6 complex in response to Wnts . DKK1 promotes the internalization and degradation of LRP6 when it forms a ternary complex with the cell surface receptor Kremen. DKK1 does not act as a head inducer during development, but also regulates joint remodeling and bone formation , suggesting a role for DKK1 in the pathogenesis of rheumatoid arthritis and multiple myeloma. Recent studies report that DKK1 affects eye development from a defined developmental time point and is essential for the separation of the lens from the surface ectoderm through β-catenin -mediated Pdgfr α and E-cadherin expression.
