background:
Folate receptor 1 (FOLR1), also known as folate receptor alpha, folate binding protein (FBP), FOLR, is a member of the folate receptor (FOLR) family. Members of this gene family have a high affinity for folic acid and several reduced folate derivatives and mediate the delivery of 5-methyltetrahydrofolate to the cell interior. Mature FOLR1 is an N-glycosylated protein anchored to the cell surface via a GPI bond. FOLR1 is primarily expressed on epithelial cells and is significantly upregulated in many cancers. FOLR1 is internalized into the endosomal system where it dissociates from the ligand before being recycled to the cell surface. Soluble forms of FOLR1 can be proteolytically shed from the cell surface into serum and breast milk. FOLR1 deficiency is the cause of neurodegeneration due to brain folate transport defect (NCFTD). NCFTD is an autosomal recessive disorder caused by brain-specific folate deficiency early in life.
