background:

B7-1 and B7-2 together with their receptors CD28 and CTLA4 constitute one of the major co-stimulatory pathways regulating T cell and B cell responses. Although both CTLA4 and CD28 can bind to the same ligands, CTLA4 has a 20- and 100- fold higher binding affinity to B71 and B72 than CD28 and is involved in the downregulation of immune responses. B lymphocyte activation antigen B7-1 (B7 for short), also known as cluster of differentiation 80 (CD80), is a member of the cell surface immunoglobulin superfamily and is expressed on activated B cells, activated T cells, macrophages, and dendritic cells. It is a ligand for two different proteins on the surface of T cells: CD28 (for autoregulation and cell-to-cell binding) and CTLA-4 (for attenuation of regulation and cell-to-cell separation). CD80 works synergistically with CD86 to provide energy to T cells. CD80 plays a role in inducing innate immune responses by activating the NF-κB signaling pathway in macrophages. Therefore, CD80 is considered an ideal target for the treatment of autoimmune diseases and various cancers.
