background:

Caspase B (CTSB), also known as APP secretase (APPS) and CPSB, is an enzyme protein belonging to the peptidase C1 family. CTSB is synthesized as a proenzyme. After removal of the signal peptide, the inactive proenzyme undergoes further modifications, including removal of the pro-region, to form the active enzyme. The catalytic activity of APPS includes: hydrolysis of proteins with broad specificity for peptide bonds; preferential cleavage of -Arg-Arg-| -Xaa bonds in small molecule substrates (thus different from cathepsin L); in addition to being an endopeptidase, it also has peptidyl dipeptidase activity that can release C-terminal dipeptides. As a thiol protease, CPSB is believed to be involved in the degradation and turnover of intracellular proteins and is also associated with tumor invasion and metastasis. Overexpression of CTSB has been associated with esophageal adenocarcinoma and other tumors.
