background:
Proprotein convertase tilisin /kexin type 9 (PCSK9), also known as NARC1 (neural apoptosis-regulating convertase), is a newly discovered subclass of peptidase that belongs to the S8 subfamily. Mouse PCSK9 is synthesized as a soluble zymogen and undergoes autocatalytic intramolecular processing in the endoplasmic reticulum, resulting in cleavage of its front peptide, which remains associated with the secretory active enzyme with a broad alkaline pH range. The protein plays a major regulatory role in cholesterol homeostasis. PCSK9 binds to the epidermal growth factor-like repeat A (EGF-A) domain of the low-density lipoprotein receptor (LDLR) and induces LDLR degradation. PCSK9 may also play a role in the differentiation of cerebral cortical neurons. Mutations in this gene are associated with a rare autosomal dominant familial hypercholesterolemia (HCHOLA3).
