background:
Recombinant human TIGIT ECD protein ( hFc tag) is a fusion protein of recombinant human TIGIT ECD (T cell immunoreceptor with Ig and ITIM domains) and human IgG1 Fc. TIGIT, also known as V-set and transmembrane domain-containing protein 3 (VSTM3) or V-set and immunoglobulin domain-containing protein 9 (VSIG9), is a novel surface protein that contains immunoglobulin variable domains, transmembrane domains, and immunoreceptor tyrosine-based inhibitory motifs (ITIMs). TIGIT is expressed on regulatory, memory, activated T cells and NK cells. It binds PVR with high affinity and PVRL2 with low affinity, but not PVRL3. Inhibition of TIGIT with siRNA in human memory T cells did not affect T cell responses, but TIGIT directly inhibited NK cell cytotoxicity through its ITIM. TIGIT inhibits T cell activation by promoting the generation of mature immunoregulatory dendritic cells. Binding of PVR to TIGIT on human dendritic cells increased IL-1 production and reduced IL-12p4 production. Furthermore, TIGIT inhibitors can suppress NK -mediated tumor cell killing and protect normal cells from NK- mediated cytotoxicity, thereby providing an “alternative self” mechanism for MHC class I inhibition.
