Thermophilic protease is a thermostable extracellular metalloendopeptidase with low substrate specificity, thus producing many short fragments suitable for sequencing. Thermophilic protease can perform limited proteolysis, preferentially cleaving at the N-terminus of hydrophobic amino acid residues, including leucine, phenylalanine, valine, isoleucine, alanine and methionine, and is used for the study of peptide localization and protein structure and conformational changes.
