Carboxypeptidase B, also known as peptidyl-L-lysine (L-arginine) hydrolase, belongs to the metal carboxypeptidase A and carboxypeptidase B (CPA/CPB) family. Each molecule contains a Zn atom and can selectively hydrolyze arginine and lysine at the carboxyl end of proteins or polypeptides. The activity is competitively inhibited by arginine and lysine, and metal ion chelators such as EDTA inhibit enzyme activity.

Rhinogen ^® Carboxypeptidase B has the same amino acid sequence as rat pancreatic carboxypeptidase B. It is a recombinant carboxypeptidase B produced by recombinant expression in E. coli and purified by chromatography. The molecular weight is 33.8kD, the isoelectric point is 6.0, and the optimum pH is 7.0-9.0. It has good enzymatic activity in the range of 25℃-37℃, and can also be digested by appropriately extending the reaction time at low temperature.

Recommended use:

1. Reagent preparation:

1) Before use, remove Rhinogen ^® Carboxypeptidase B and centrifuge at 10,000 rpm for 10 seconds to ensure that all the dry powder is at the bottom of the tube; dissolve the lyophilized powder with deionized water or 10mM Tris-HCl (pH7.5) to make the enzyme solution concentration 1-10mg/ml.

2. The recommended usage of Carboxypeptidase B protein is, enzyme amount: target protein (mass ratio) = 1:50~1:1000 (w/w), the optimal pH is 7.5-9.0, and the enzyme digestion temperature is 25℃-37℃.

Notice:

1) For different protein samples, it is necessary to experiment to find the most appropriate enzyme concentration and reaction time.

2) Reaction at 25℃-37℃. When digesting at 25℃, the reaction time can be appropriately extended.

3) The optimal pH of the enzyme digestion buffer is 7.5-9.0. When the pH is ≥12, the activity is completely lost.
