Overview:
α-2,6-Sialyltransferase 1, also known as ST6GAL1, is responsible for facilitating the transfer of sialic acid from CMP-sialic acid to galactose-containing acceptor substrates. Galβ-α-2,6Sialyltransferase 1 (β-galactosamine-α-2,6-sialyltransferase 1), encoded by the ST6GAL1 gene, is a type II membrane protein located in the trans-Golgi network that catalyzes the 2,6-sialylation of β-galactose-1,4-N-acetyl-D-glucosamine (Galβ-1,4-GlcNAc) structures on N-glycans. Sialyl molecules attached to glycoproteins or glycosphingolipids play an important role in various biological processes such as immune recognition, pathogen infection, and cell adhesion. Sialyltransferases are key enzymes that regulate the cellular levels of sialic acid-containing molecules. The enzyme is involved in the generation of cell surface carbohydrate determinants and differentiation antigens HB6, CD75, and CD76.
Enzyme activity definition :
α-2,6-Sialyltransferase 1 transfers N-acetylneuraminic acid (Neu5Ac) from CMP-sialic acid (CMP-Neu5Ac) to N-acetyllactosamine (N- Acetyllactosamine ). Enzyme activity is defined by measuring the amount of CMP. Under the conditions described (25 mM Tris, 150 mM NaCl, 5 mM CaCl 2 , 10 mM MnCl 2 , pH 7.5), 1 μg of enzyme can release > 150 pmol of CMP per minute .
