background:
Fucose exists in different linkages on N- and O-glycans and is an important component of many oligosaccharide (O- or N-linked) and polysaccharide structures, as well as glycoproteins and glycolipids, which are often involved in various biological events, including fertilization, embryogenesis, apoptosis, signal transduction, and disease progression, such as rheumatoid arthritis, inflammation, cancer, and cystic fibrosis.
Rhinogen ® α1-3,4 fucosidase is recombinantly expressed in E. coli. It has an optimum pH of 6.0 and a working pH range between 5.5 and 7, within which the enzyme maintains at least 50% of its maximal activity. The enzyme efficiently recognizes α1-3,4 fucosylated glycans (e.g. Lewis X/A epitopes, including their sialylated counterparts) and hydrolyzes terminal α1-3 and α1-4 fucosyl bonds in these substrates without removing the sialic acid moiety. Since no sialidase is required prior to targeted digestion of antennary fucosylation, this enzyme can be used to simplify the workflow, making it both economical and time-saving. Another advantage is that the enzyme can be used to monitor α1-3,4 fucosylation on both free glycans and glycoproteins, eliminating the need for chemical or enzymatic release of glycans from glycoproteins.

Recombinantly expressed in E. coli , with a theoretical molecular weight of approximately 62kDa and a 6×His tag at the C-terminus .
Product quality:
SDS-PAGE analysis showed that the purity was ≥95%; no contaminating exoglycosidase, endoglycosidase and protease activities were detected.
Enzyme activity definition :
hydrolyzes ≥90% of fucose from 4.5 nmol 3'-fucosyllactose when incubated in 50 mM sodium acetate, pH 6.0 at 37°C for 30 min.
Storage conditions:
1. Use ice packs for transportation. Please place the enzyme at 2-8℃ immediately after receiving the product; the supporting reagents should be stored at -20℃.
