background:
EndoS is a highly specific IgG endoglycosidase that hydrolyzes and cleaves the β-1,4 glycosidic bond in the middle of the chitobiose core of the N-glycan on the IgG Fc segment , releasing the sugar chain and leaving GlcNAc (N-acetylglucosamine) with or without core fucose on the Asn residue of the glycoprotein .
Rhinogen ® EndoS endoglycosidase is a recombinant EndoS glycosidase produced by E. coli expression system and purified by multi-step chromatography . It contains a His tag, which can be easily separated and removed from the enzyme digestion reaction system after the digestion is completed. It can effectively digest all types of N- glycans , including high mannose, hybrid and complex types. It can act on all human IgG, as well as mouse, rat, monkey, goat, sheep, cattle and horse IgG antibody sugars.

Rhinogen ® EndoS endoglycosidase is a recombinant EndoS endoglycosidase produced by recombinant E. coli expression system and purified by multi-step chromatography . Its molecular weight is about 92kDa.

The optimum reaction pH is 6.0~8.0; the optimum reaction temperature is 37℃.
Enzyme activity definition :
Definition of 1 unit of enzyme activity: the amount of enzyme required to digest ≥95% of 1 μg of recombinant monoclonal IgG within 30 minutes at 37°C.
Storage conditions:
Use ice packs or dry ice for transportation. Please freeze the product at -30℃ to -10℃ immediately after receiving it. Avoid repeated freezing and thawing.

application:
Deglycosylation to remove Fc glycan heterogeneity; antibody glycoengineering ; IgG glycoform analysis .
Recommended use:
1. Before use, please put Rhinogen ® Take out the EndoS endoglycosidase and centrifuge at 10,000 rpm for 10 seconds to ensure that all the enzyme solution is at the bottom of the tube;
2. Add the required amount of IgG to the digestion buffer (the recommended IgG concentration is 0.5~10mg/ml);
Note: The recommended digestion buffer is 10mM sodium phosphate, 150mM NaCl, pH 7.4;
3. Add Rhinogen ® EndoS endoglycosidase was added to the IgG sample;
Note: Add 1 unit of EndoS glycosidase for every 1 μg of IgG for digestion; for example, add 1 μl (40 units) of EndoS solution to digest 40 μg of IgG;
4. Incubate at 37°C for 30 minutes.
